Abstract

Applying of biotechnology is very important in plant breeding, especially to conservation goals. The aim of this research is to conservation of Tarusan mango by in vitro technique. This research consist of two research phase, that is initiation phase and multiplication Phase. At initiation phase, conducted examination to 3 basal medium (MS, WPM,B5) and also antioksidan (Ascorbic Acid and Chorcoal), to get the establish of medium and antioksidan to supporting initiation of embryo culture of Tarusan mango. At phase of multiplication conducted examination of combination plant regulator of NAA ( 0.0,1.0,2.0, and 3.0 ppm) and Kinetin (0.0, 2.0, 4.0, and 6.0 ppm). Result of this research is, in initiation phase, the role of 3 basal medium (MS, WPM and B5) can support to embryo culture initiation of Tarusan mango. B5 basal medium more influence to quickly the explant to form the rootlet (9.53 DAT) and shootlet (17.70 DAT). For the treatment of antioksidan, the charcoal is better influence to percentage of explant life (75.24%), and also the percentage of explant to forming the shootlet (62.16%). At multiplication phase, the treatment of plant regulator combination of NAA-KINETIN give influence of interaction to percentage of explant life. The combination of 1.00 ppm NAA + 2.00 ppm more support the highest percentage of explant life than another combinations (41.28%). The concentration of NAA (higher level than Kinetin (3.00 ppm NAA + 0.00 ppm Kinetin; 3.00 ppm NAA + 2.00 ppm Kinetin) can induce the explant to form the rootlet for epicotyl culture. The contrary, when concentration of Kinetin is higher than NAA concentration (0.00 ppm NAA + 2.00 ppm Kinetin) can induce the explant to form the shootlet for epicotyl culture.
