Abstract
This study aimed to observe the embryos viability of Pesisir cattle were biopsied in the implementation of the embryo sexing (determining the sex of the embryo), and to determine the effectiveness of the determination of the sex of Pesisir cattle embryos using PCR and to investigate the sex ratio in morula and blastula phases using 1 and 2 blastomeres. It is expected that this research contributes to the development of reproductive biotechnology cattle which the born of male or female cattle have an uniformity depending on the expected population structure.This research was conducted in UPT of Animal Science Faculty University of Andalas (Superovulation), Laboratory of Animal Reproduction at Animal Science Faculty, Andalas University (embryo evaluation), and Laboratory of Embryology at Faculty of Veterinary Medicine IPB (embryo biopsy and determination of the sex of the embryo ). This research was done between 1st January 2012 and 30th September 2012.The materials were Pesisir cattle embryos obtained from the superovulation of cows and bulls that older than 3 years, and the body weight about 140.69 ± 5.51 kg. Used 15 embryos of Pesisir cattle that have superovulation using CIDR and FSH hormones, an then the embryos were biopsied with microblade the morula and blastula phases by taking 1 and 2 blastomeres. The variables were the embryo viability at 1,3,6 and 12 hours after the biopsy. The effectiveness of sex determination using PCR with primers BOV 97m and sex ratio of the embryo. Data were analyzed using the chi-square test and Fisher's test.The results of this study stated that the embryo viability at the morula phase used biopsy 1 cell and 2 cells for 1 hour, 3 hours, 6 hours and 12 hours was the same, whereas the embryo viability used biopsy 1 cell and 2 cells at the blastula phase for 1 hour, 3 hours and 12 hours of embryo viability was the same but with biopsy 2 cells at the blastula phase where the 6 hours after the biopsy that the progression was better when compared to both embryo viability after biopsy like biopsy 1 and 2 cells at the morula phase and biopsy 2 cells at the blastula phase. Meanwhile, the statistically analyzed using the Fisher test that the embryo viability for 6 hours after the biopsy at the morula and blastula phases with 1 cell and 2 cells was not significant (P > 0.05). The effectiveness of embryo sex determination in morula phase using 1 cell and 2 cells was same (100%), in blastula phase using 1 cell was 91.67% and using the 2 cells was 100%. After statistically tested using Fisher test showed that the effectiveness of sex determination in the blastula phase using 1 cell and 2 cell was not significant (P > 0.05). Sex ratio of embryos at the morula phase using 1 cell and 2 cells was the same that for the embryo male sex about 66.67% and embryo female sex about 33.33% while at the blastula using 1 blastomer showed the male sex ratio around 41.67% and the female sex embryo was 50%, using 2 blastomeres showed that male sex ratio was 41.67% and the female sex was 58.33%. Sex ratio of embryos at the blastula phase using 1 cell and 2 cell after statistically tested using Fisher test were not significantly (P > 0.05).
