Abstract
The objective of this study were (1) to identify polymorphism of growth hormone gene and to characterize nucleotide changes and its position in the DNA sequence from exon three to 3’ flanking region in Pesisir cattle (2) to determine the association between the polymorphism of growth hormone gene and body weight in Pesisir cattle. The body weight of Pesisir cattle showed a high variability. This finding study suggested the body weight can improves through selection. The sequenced data from four PCR product showed high variability in bGH gene of Pesisir cattle. Comparing with NCBI GenBank Acess M57464.1. From four fragmen 18 deletions, 15 insertions, and 21 mutations were identified. Of the 18 bGH gene deletion in the Pesisir cattle 6 are specific to these cattle because it occurs at all samples and not yet found in other seqeunce in NCBI GenBank, that are deletion of A at position 1408, deletion of G at position 1410, deletion of T at position 1412 , deletion of C at position 1414/1415, deletion of A at position 1429, and the deletion of T at position 2432. Of the 18 deletions 10 of them are polymorphic and 8 others are monomorphic. Of the 15 insertions in the bGH gene Pesisir cattle, 2 of which were specific to these cattle since it occurs at all samples and not yet found in other bGH sequence in the NCBI GenBank, that are G insertion at position 1549-1551, C insertion at position 1895. From 15 insertion 4 of which are polymorphic and 11 are monomorphic. Of the 21 mutations in the bGH gene Pesisir cattle 3 of which were specific to Pesisir cattle since it occurs at all samples and not yet found in other bGH sequence in the NCBI GenBank. The mutation are T G mutation at position 1947, C G mutation at position 2639, C A mutation at position 2813. Of the 21 mutations are 17 of them are polymorphic and the other 4 are monomorphic. The results of the χ2 analysis showed no relationship between bGH gene polymorphisms and two weight groups of Pesisir cattle.
