Abstract
Beef meatballs as a food favored by the people of Indonesia are prone to pork contamination because beef is relatively expensive. The mixing case certainly causes inconvenience for Muslim communities in Indonesia because pork is not halal to be consumed. However, it was necessary to analyze pork contamination in beef meatballs. Samples were taken from the Tanjung Priok area because the number of adherents who could consume pork in the area was quite significant. Three control meatballs (100% pork, 100% beef, 50% pork) and 10 sample meatballs were analyzed using real-time polymerase chain reaction (RT-PCR) method. The DNA isolate used was mitochondrial DNA with a cytochrome B target area. The results of RT-PCR amplification in 3 control meatballs using pork primer showed 100% pork meatballs and a mixture of 50% pork resulting in an increase in the amplification curve with crossing point (CP) 22.82 and 20.03. While the results of the amplification of 10 samples of beef meatballs using pork primers did not increase the amplification curve which showed that there was no amplified porcine DNA in the beef meatball products
