Abstract

Experiment on the isolation of IAA producing bacteria from acid soil and moleculer identification have been undertaken in the laboratory of biotechnology and plant breeding faculty of agriculture, andalas university in padang. The experiment was conducted from may 2009 until may 2010. The purpose of this study were to obtain bacteria isolate capable of producing high IAA from acid soils and moleculer identification using 16S Rrna gene. This experiment concicted of 2 stages : (i) Identification and characterization of bacteria in morphology and molecular. The soil samples were taken from the area around campus experiment station Unand Limau Manih on some rhizosfir crops such as a Alang-alang (Imperata cylindrica), Sikaduduk (Melastoma malabathricum), Karamunting (Rhodomyrtus tomentose), Paku resam (Dicranopteris linearis), Pimping (Themeda gigantea) and Polysticum sp. Bacteria isolate that could produce high IAA were subjected to the next molecular identification using 16S Rrna GENE. 50 bacterial isolates were selected based on shape and color of the colony. Isolate alang-alang-10 known to have the ability to produce highest IAA compared to other isolates. That’s why the bacterial isolates alang-alang-10 was selected for further molecular characterization using 16S Rrna gene. Based on BLAST analysis showed that isolates alang-alang- 10 is a bacterium of the genus acinetobacter. Key words : bacteria, IAA, Acid soil, 16S Rrna gene
