Abstract

This experiment sets out to examine the effect of rilmenidine on blood pressure (Systolic blood pressure = SBP, Diastolic blood pressure SBP) reduction and heart rate (HR) on acute saline volume expansion Sparague-Dawley (SD) male rats. The animal were anaesthetized with a mixture of fluothane/O2¬ and thereafter maintained on chloralose/urethane administrated i.v.via femoral vein. Changes in SBP, DBP and HR were monitored using a Grass transducer connected to LabVIEW data acquisition system on a Macintosh computer. Saline (0.9% w/v NaCl) was infused at 3 ml/l and the experiment begun 2h later. Following the collection of baseline data, bolus doses of rilmenidine, 0, 100, 300 and 1000 µg/kg body weight were administered 15 minutes before saline infusion was expanded, such that the animal received 0.25% body weight/min over 40 min, and block of recording were made every 5 min. the data obtained was compared using 2 way ANOVA and was considered to be significant when P,0.05. The result showed that, saline volume expansion on the SD rats reduced the lowering blood and HR of rilmenidine. Keywords : Rilmenidine, Blood Pressure and Saline Volume expansion
